Multicellular Feedback Control of a Genetic Toggle-Switch in Microbial Consortia

چکیده

We describe a multicellular approach to control target cell population endowed with bistable toggle-switch. The idea is engineer synthetic microbial consortium consisting of three different populations. In such consortium, two populations, the Togglers, responding some reference input, can induce switch memory mechanism in third population, Targets, so as activate or deactivate additional functionalities cells. Communication among populations established by orthogonal quorum sensing molecules that are used close feedback loop across design validated via in-silico experiments BSim, realistic agent-based simulator bacterial

Noise in Genetic Toggle Switch Models

In this paper we study the intrinsic noise effect on the switching behavior of a simple genetic circuit corresponding to the genetic toggle switch model. The numerical results obtained from a noisy mean-field model are compared to those obtained from the stochastic Gillespie simulation of the corresponding system of chemical reactions. Our results show that by using a two step reaction approach...

Genetic toggle switch without cooperative binding.

Genetic switch systems with mutual repression of two transcription factors are studied using deterministic and stochastic methods. Numerous studies have concluded that cooperative binding is a necessary condition for the emergence of bistability in these systems. Here we show that, for a range of biologically relevant conditions, a suitable combination of network structure and stochastic effect...

The Switch in a Genetic Toggle System with Lévy Noise

A bistable toggle switch is a paradigmatic model in the field of biology. The dynamics of the system induced by Gaussian noise has been intensively investigated, but Gaussian noise cannot incorporate large bursts typically occurring in real experiments. This paper aims to examine effects of variations from one protein imposed by a non-Gaussian Lévy noise, which is able to describe even large ju...

A cholesterol toggle switch.

Cholesterol levels in mammalian cells are controlled by an intricate mechanism in which the transcription factor SREBP plays a key role. Work in this issue (Radhakrishnan et al., 2008) employing direct measurement of endoplasmic reticulum cholesterol levels offers insights into the "switch" that controls this system with surprising precision.

Biological Signal Processing with a Genetic Toggle Switch

Complex gene regulation requires responses that depend not only on the current levels of input signals but also on signals received in the past. In digital electronics, logic circuits with this property are referred to as sequential logic, in contrast to the simpler combinatorial logic without such internal memory. In molecular biology, memory is implemented in various forms such as biochemical...
